DC SIGNR silencing reduces HIV-1 infection in Dendritic cells
نویسندگان
چکیده
Results High expression of DC SIGN/R was observed on DCs infected with HIV-1 (p= 0.01). DC sIGNR expression in DCs was maximally downregulated by siRNA at 24 hrs (p=0.002) and was associated with significant reduction in expression of CD40 (p= 0.003), CD80 (p=0.008), CD86 (p=0.007) and P38 MAPK (p=0.005). Transfection of DC SIGNR on DCs at 24 hours followed by infection with clade C HIV-1 demonstrated lower levels of p24 compared to that in untransfected DCs (p=0.0008).

In 1716 individuals--801 human immunodeficiency virus (HIV)-1-seropositive individuals, 217 high-risk HIV-1-seronegative individuals, and 698 general HIV-1-seronegative individuals--from a Seattle cohort and a Multicenter AIDS Cohort Study cohort, the association between HIV-1 susceptibility and repeat-region polymorphisms in the gene for the dendritic cell-specific intercellular adhesion molec...
متن کاملDC-SIGN (dendritic cell-specific ICAM-grabbing non-integrin) and DC-SIGN-related (DC-SIGNR): friend or foe?
C-type lectins are calcium-dependent carbohydrate-binding proteins with a wide range of biological functions, many of which are related to immunity. DC-SIGN (dendritic cell-specific ICAM-grabbing non-integrin, where ICAM is intercellular adhesion molecule) is a recently described mannose-specific C-type lectin expressed by dendritic cells. Dendritic cells are potent antigen-presenting cells cap...
متن کاملHepatitis C virus glycoproteins interact with DC-SIGN and DC-SIGNR.
DC-SIGN and DC-SIGNR are two closely related membrane-associated C-type lectins that bind human immunodeficiency virus (HIV) envelope glycoprotein with high affinity. Binding of HIV to cells expressing DC-SIGN or DC-SIGNR can enhance the efficiency of infection of cells coexpressing the specific HIV receptors. DC-SIGN is expressed on some dendritic cells, while DC-SIGNR is localized to certain ...
متن کاملFunctional Genetic Variants in DC-SIGNR Are Associated with Mother-to-Child Transmission of HIV-1
BACKGROUND Mother-to-child transmission (MTCT) is the main cause of HIV-1 infection in children worldwide. Given that the C-type lectin receptor, dendritic cell-specific ICAM-grabbing non-integrin-related (DC-SIGNR, also known as CD209L or liver/lymph node-specific ICAM-grabbing non-integrin (L-SIGN)), can interact with pathogens including HIV-1 and is expressed at the maternal-fetal interface,...
متن کاملStructural basis for selective recognition of oligosaccharides by DC-SIGN and DC-SIGNR.
Dendritic cell specific intracellular adhesion molecule-3 (ICAM-3) grabbing nonintegrin (DC-SIGN), a C-type lectin present on the surface of dendritic cells, mediates the initial interaction of dendritic cells with T cells by binding to ICAM-3. DC-SIGN and DC-SIGNR, a related receptor found on the endothelium of liver sinusoids, placental capillaries, and lymph nodes, bind to oligosaccharides t...
